I heard of someone a while back that had attempted to replicate something similar to 69Ron's SWIM's work with calamus.
20g calamus was shredded and washed in 400ml acetone for 6 hours, the acetone was filtered and discarded, and the mush was allowed to dry thoroughly. The washed and dried mush was added to 400ml distilled, room temperature water and soaked overnight, filtered and discarded to produce approx 400ml of tea. The tea was then divided into two halfpint mason jars. To find the active dose and test for allergies and side effects, the plan was to ingest 1/3 of one jar at one sitting, then the second test would be the rest of the first jar (2/3), then the third test would be the entire second jar (3/3).
The first attempt had no effects, but also no side effects. At the time of the second dose, further reading showed that Ron's SWIM had gone much stronger with additional calamus, and it was decided in the moment that the amount remaining would be perfectly reasonable, so down the hatch it went. At worst there was a mild stomachache that was easily remedied with a small snack, at best there was a pleasant relaxation for a few hours. Hard to say how accurate the notes are, but theoretically then, that last dose was a tea of approx 17g calamus washed with an excess of (maybe too much?) acetone and then extracted with water. No psychedelic effects of any kind were perceived, and as far as I know no further attempts were made, but I will keep my eyes and ears open.
To respond to your questions; 1. I would think fresh is better but I could be wrong. basically it's "old tea". Oxidation could reduce the effects, but could also catalyze them I suppose. 2. Everyone is different, so it is always wiser to start slow and move up, and in doing so titrate your own tolerance, rather than making wholly uneducated guesses. 3. No idea about powdered extracts or other methods of ingestion. Try it and report back? 4. The active chemicals seem to be a point of contention last I checked, hard to say exactly what you would be looking to purify. but as you say you are well-equipped, proceed with caution and please share whatever useful knowledge you can derive.
Good luck! Sorry I couldn't be of more help than that.

....pretty much unsurprisingly. I may also add. The rationale of doing these steps would be what??? That the trippy actives in calamus are alkaloids? Wrong assumption here. "Active principle" does not equal "alkaloid". Need to calculate between salts and freebases? Click here! Need to calculate freebase or salt percentage at a given pH? Click here!

The actives in calamus you want is not asarone, but something different. I believe it is not an alkaloid, but rather a phenethylamine-like substance without nitrogen. Asarone does suit this, but it is not psychedelic. Many related substances are also found in calamus, I think one of these is the active. It may also be that an unidentified compound is responsible. The actives is not alpha- or beta-asarone, that's for sure. The psychedelic substance is soluble in cold water. It seems like acetone is pulling out the shit that makes you purge so bad, I think asarone is the responsible. You should check out this thread from 69ron, it has very much interesting information. I wish you good luck, can you keep us updated on your work? I have a feeling that it is an entheogenic treasure chest waiting to get opened inside of calamus.

Ive seen a number off documentaries and so forth regarding DMT and the effects and processes I've also been intrigued on information about how its traced usable up to 2mg per hundred grams in terms of it being evidently enough to get a Journey from.
have some queries one about if anyone has Heard of Angels Tears its been a term to describe acacia tree sap and high amounts of DMT and how would one extract DMT from this naturally our Bush Masters wouldn't have had Refined chemicals surely " the only one that stands out if you wash down certain parts of trees for potassium purity before it becomes a metal.
also smoking it wouldn't need the MOI

also smoking it wouldn't need the MOI I think you've posted in the wrong thread. Maybe the wrong universe Seriously, what is "the only one that stands out if you wash down certain parts of trees for potassium purity before it becomes a metal" even supposed to mean? It's like one of those bot posts on Yxxtube comments. Welcome to the Nexus, btw. Just how high were you when you posted this? “There is a way of manipulating matter and energy so as to produce what modern scientists call 'a field of force'. The field acts on the observer and puts him in a privileged position vis-à-vis the universe. From this position he has access to the realities which are ordinarily hidden from us by time and space, matter and energy. This is what we call the Great Work." ― Jacques Bergier, quoting Fulcanelli
