Just posted a bit about me in my introduction essay. I have been wondering something for sometime that I can't seem to find the answer to, although my gut tells me I found the answer. Maybe you can help...

I have heard great things concerning both Huperzine and Datura S. regarding synergy and potentiation of cactus, in specific mescaline. I was wondering, does Huperzine and Datura S. cancel each others effect?

From my research, the main component of Datura S. is Hyoscyamine as well as smaller amounts of Scopolamine and Atropine. Hyoscyamine is an antagonist of muscarinic acetylcholine receptors while Huperzine is reversible acetylcholinesterase inhibitor, inhibiting the breakdown of the neurotransmitter acetylcholine by the enzyme acetylcholinesterase.

At first glance it appears that one would cancel out there other, but Hyoscyamine blocks the action of acetylcholine at parasympathetic sites in sweat glands, salivary glands, stomach secretions, heart muscle, sinoatrial node, smooth muscle in the gastrointestinal tract, and the central nervous system. I am not sure if Hyoscyamine also black the effect of acetylcholine in the CNS as well.

If it doesn't affect CNS, is it possible there could be enhanced synergy of these two compounds when taken with mescaline? Is there a safety issue?

What have you heard about the synergy of these two things with mescaline?

I've heard about datura being added in low doses to cactus tea, primarily because of it nausea-suppressing properties. But I've never even heard of Huperzine before.

Have you heard that these substances will potentiate the mescaline? Its been my experience that cannabis potentiates mescaline nicely.

There are horror stories about datura... that people harm themselves or get badly injured without realizing it while intoxicated. Of course the effects are dosage dependent, but be careful with datura because it can be dangerous.

Hey Greyfox, thanks for the welcome.

I have heard from 69ron that low dose Datura S. (1-5 seeds) does take nausea, but also that it increases the strength of mescaline when mescaline is also taken in low doses which is where I find a lot of value. He claimed that it could almost double the potency when mescaline is in the lower dosage range but when mescaline is at higher dosage it tends to have negligible effect, albeit still taking away nausea some.

Also, I appreciate the concern regarding the safety of Datura. I understand the risks associated with higher usage and thus have much respect and caution when dealing with it.

Huperzine A is a naturally derived reversible acetylcholinesterase inhibitor and NMDA receptor antagonist. It is works in the same manner as currently approved FDA treatments for Alzheimer's such as Galantamine and Donepezil. People often use it for lucid dreaming with success. I don't find it stimulating. I find it very beneficial on its own for studying and learning new things, retaining information, language learning, etc. It also helps bring about a clarity that is useful when meditating. Dosage is in the micrograms of around 50-200 for the average person. It must not be used for long periods of time. I never take it two days in a row. I usually take every 4-6 days, giving my choline a chance to recuperate.

The experience report I read said that Huperzine A brought an increased clarity to the experience. They also said it sharpened the visuals and potentiated the mescaline some as well.

Hyoscyamine certainly does block the effect (or more accurately, act as an inverse agonist) of acetylcholine in the CNS, which is evident by it's sedating effects and in higher dosages, memory loss, delirium and hallucinations in overdose.

Taking hyoscyamine and huperazine a together is seemingly counterproductive to me as well, since acetylcholinesterase inhibitors are actually able to reverse the effects of anticholinergic overdose.

As for the synergy with mescaline, I'm not sure, it would depend what you are looking for. Huperazine A, or generally acetylcholinesterase inhibitors or other agents that increase the effect of acetylcholine, (piracetam, noopept) are used as nootropic agents for increased concentration and focus, this could be combined with a microdose of mescaline.

In the past actually i've taken a similar combination, huperazine a, oxiracetam, vinpocetine, a low dose of mescaline sulfate (50mg) with some coffee.

I didn't find the combination particularly useful, too much stimulation. I find mescaline alone is stimulating enough, and not very useful as a microdose, because it becomes difficult to focus.

However, for psychedelic doses of mescaline, I can certainly see the benefit in huperazine a adding certain clarity to the experience, like described above.
But remember, there is always a trade off. I find when I take cholinergic agents, my mind is sharper, and more analytical. However the excess of cholinergic activity tends to blunt my emotions, which is not necessarily something you want during a psychedelic experience. They also tend to make me more irritable.

I wouldn't use atropine or hysocamine for the same purpose, at all. Those compounds I sooner would use if I wanted sedation to help sleep, after a mescaline experience.

Thank you for the insight. That seems like a decent amount of nootropics to add with something as stimulating as mescaline indeed. Never taken Oxiracetam but have heard it can cause excess stimulation as well as some anxiety/nervousness. Definitely something to be avoided in these situations.

I guess Huperzine might be helpful at the lower-microdose level for its ability to focus as you mentioned experiencing a loss in focus at these doses. I also notice something similar. I think in my case, there is a little loss of focus, but more so, the task I want to focus on changes, meaning my interests shift. For instance, if I am at work I will find myself doing some research on something personal to me rather than work related. Possibly throwing on some good tunes. In these scenarios, my focus seems pretty darn good. haha

That said, it seems like Huperzine may be useful at higher doses to help with the integration process since it seems to help with memory and analytics so well. Although there is always a tradeoff as you mentioned some emotional blunting. Definitely something to consider. Thanks for the input
