I think that many of you have already heard this.
"If you don't get your dose in 40 seconds (or something like that), tolerance will quickly build up, and DMT won't have any effect anymore, no matter how much you take after that".
What does your experience tell you about this? Is this false or true?

True in my experience, at least when talking about breakthrough doses. Usually the intensity of the experience is inversely proportional to the time it takes to ingest the material. If you're not fast enough, often it can lead to an aborted breakthrough.
For sub-breakthrough doses, I think the behavior is different. You can continue to vape more DMT, and the intensity of effects will increase, but you will not breakthrough. Often trying too hard too late will result in the sensation of internal head pressure or a trip that goes nowhere.
Edit: things are totally different when you add harmalas to the mix, then the doses tend to stack.

I'd say mostly true. I've had experiences where I have sucked in a ridiculous (looking back) amount of vapor within a two minute period and got nothing from it. Other times it hit me right away after the first little hit. No matter my method it's always a little unpredictable, but with my super-bong-health-stone method it's been easier. I'd say that for my deepest journeys the whole inhalation and holding process for the multiple hits took about 40 seconds total. Making sure you're vaporizing rather than burning, taking huge lungfuls, and holding it in for at least 10 seconds are the most important components however. My most powerful, deepest DMT journeys have been the ones where I inhaled the whole vape in one big lung-full & held it for at least 10 seconds. That is not to discount the fact that I've had pleasurable, albeit non-breakthrough trips taking small inhalations many times over a few minute period. I would say true. If I smoke changa and after about lets say 15-20 minutes I take same dose again I feel nothing at all. If I wait longer one hour or more then I can start smoking again. I hear some people can smoke changa for hours without break but it would not work for me. We are each of us angels with only one wing, and we can only fly by embracing one another.

Interesting. I see only one who says it isn't, and all other who say it's true. Thanks. I didn't have enough time to experiment, but I think that one time I inhaled ridiculous amount during minute or more, and I had sub breakthrough experience. Looks like first two hits are most important, and should be done rapidly.
