Regarding your hefty build, I don't think it has an affect on aya or DMT dosing and minor effect on LSD dosing. I presume other members will chime in if they know differently but as far as I can tell, that's the case.
Most cross-tolerance in psychedelic materials can be obliterated by a week or two of complete abstention I have found. Regardless, I would recommend starting low. You know what they say, . . if you underdose you can always take more but if you overdose your options other than to ride it out are obliterated.
50 grams of aya (banisteriopsis caapi) & 50 grams of chacruna sounds like a very nice brew to me - a hearty handshake so to speak.
But I am quite concerned about your intention to use 5 grams of MHRB (mimosa hostillis root bark). I believe Ott and others have determined MHRB to contain total of upwards of 3.0 percent NNDMT. Thus, if you were to achieve complete MAO inhibition, 5 grams of MHRB could conceivably be equivalent to 150mgs of freebase NNDMT. In my humble (& remembering horror shows and ass kickings on MUCH lower oral doses) opinion that is WAY TOO MUCH for an initial trip.
If you are going to use MHRB, probably skip the chacruna and only begin with a gram or two.
Be sure to brew enough for more than one dose. Then you can take more MAOI and/or DMT containing plant later if you want or if you purge early, etc.

Yes, hefty oral doses can approach and/or cause a breakthrough. Be careful with this though, I would recommend doing a freebase extraction with your MHRB and smoke a few breakthrough doses first. Spend 5 minutes in hyperspace before getting committed to spending 1-4 hours there . . .
