Methotrexate is a very useful, but potentially very toxic drug, whose mechanism of action is to interfere with folate metabolism by competing with an enzyme called Dihydrofolate Reductase in a competitive fashion.This has effects on DNA/RNA synthesis.
The dosing regime is usually once a week orally for conditions such as psoriasis and is relatively speaking a low dose compared to use in hematological malignancies.Psoriatic treatment doses have a half life of upto 10 or so hours (but the pharmokinetics are variable at higher doses).
Interestingly LSD has been shown to have potent inhibitory effects on inflammation mediated by a cytokine called TNF-alpha (Tumour Necrosis Factor); in people with psoriatic arthritis in whom methotrexate has failed, the next step is to use inhibitors of this cytokine.
Im not suggesting that one should use LSD to treat psoriasis (but its an interesting proposition!
).Methotrexate has no adverse interaction with ergotamine/ergometrine which are probably the closest prescribed meds in terms of structure to LSD, and with the half-life being 10ish hours at the low once-weekly regimes commonly used, I would personally take the acid 5-6 days after the methotrexate.
