I am doing a simple hermala FB extraction, skiping the doble filtration with ice and after that freebase the alcaloides.I go directly with the amonia for FB harmala after collection one purification is done redisolving the harmalas in vinager ,tilter the liquid to red dark transperent tee and one more time the amonia.I get for second time a very low yeld. And I don't think the provider's seeds are low quality.
Procedure
150 grams grinded seeds was boiled with some vinager three times for about 40 min each boil.Done at almost full speed boiling.Recolected all the liquid (very dense in color),and used a half of a bottle of amonia to change the color completly.
Weited 8h,filtered the harmalas.
Redisloved them in water/vinager and filtered the liquid.Transperent tee obtained
One more time used amonia until the liquid stop changing color.
Some gold harmala start to foll down.
Toatal ammount after filtering and drying -1 gr.
Where is the problem?

How much water used for each fraction?

As I am trying to use as little liquid as possible,I used just the amlunt to cover the seeds, anyway the recolected liquids was very dense in collor.

Not the way to go.
1. Do not grind the seeds. Harmalas are in seed shells, the kernel contains plant matter that you dont want.
2. Do not boil, only simmer on low flame. More heat - more contaminants pass into solution
3. Do not exceed 50ml/1L of vinegar. The lower the PH the more contaminants cross over. (Ideal PH is around 5.5)
4. Simmer for 40 mins. Do not reduce the liquid.
5. Preferably use sodium carbonate instead of ammonia for collection fractions. Use ammonia only for the final yield (Cost-wise). About 25g sodium carbonate per fraction. Let it cool a bit before adding carbonate.
6. Optionally, you could simmer your seeds in basic water (25g sodium carbonate per liter) for about 40 mins, then discard this fraction and wash the seeds with tap water. This improves the purity
7. Consider doing the manske step (twice in succession) to remove unwanted alkaloids.
